7-hydroxymitragynine (7-OH) is the most potent naturally occurring opioid-receptor-active compound found in any plant. A 2016 study in the Journal of Medicinal Chemistry found 7-OH is approximately 13 times more potent than morphine at mu-opioid receptors in binding assays. For context: morphine is the reference standard of opioid potency. Fentanyl is 50–100x more potent than morphine. 7-OH sits between morphine and fentanyl in receptor binding potency. However, 7-OH naturally occurs at less than 2% of total kratom alkaloids, so standard kratom powder delivers very small absolute doses of 7-OH.
